Enzymes(효소)

효소(Enzymes)




생명을 유지하는 과정에 있어서 수천가지의 신진 대사에 반응하는 생체분자 이다. 

그들은 음식의 소화에서 DNA 합성까지, 대사 반응의 속도와 특징을 가속화하는 촉매제 이다.

일부 촉매인 RNA 분자 가 확인 되고는 있지만 대부분의 효소는 단백질이다.


효소의 촉매 작용 을 지원하기 위해 공동인자인

유기(예 : 비타민 B 복합체 )를 무기( 예: 마그네슘 이온 ) 을 사용할  수 있다.


거의 모든 생물 세포의 화학 반응들은 충분한 생명력의 발생을 위해 효소를 필요로 한다.


대부분의 효소반응 속도는 비촉매 반응에 비해 수백만배 빠른 속도로 반응한다.


모든 촉매 와 마찬가지로 , 효소들은 촉매나  반응에 의해 변경되거나 소모되지 않는다.

그러나 효소는 그들의 기질이 매우 구체적이라는 점에서 대부분의 다른 촉매와 다른점이다.

효소는 4,000개의 바이오 화학 반응에 대해 촉매작용을 하는 것으로 알려져 있다. 


효소 활성은 다른 분자에 의해 영향을 받을 수 있는데,

효소 억제제는 효소활성을 감소하는 분자이고,

효소활성제는 효소활동을 증가하는 분자를 말한다. 

많은 약물과  독극물은 효소 억제제이다. 


효소활성은 온도, 압력, 화학적 환경 (예, PH), 및 배양기의 농도에 의해 영향을 받는다. 


일부 효소는 상업적으로 사용되는데, 

예를 들면, 항생제의 합성 또는 

생필품등의 생화학 반응 (분말세척제에 단백질이나 지방 얼룩을 분해),

식품 단백질 분해제(고기 단백질을 더 작은 분자로 분해하여 부드럽게 씹히게 하기위해)로 효소를 쓴다.


조효소(Coenzymes) 는 하나의 효소에서 다른효소로 화학물질을 운송하는데,

리보플라빈 , 티아민 및 엽산 등과 같은 비타민(신체에 의해 합성 될 수 없어, 반드시 식단에서 취득해야 화합물)등이 

바로 그런것들이다.


보통 조효소(Coenzymes)는 세포 내부에서 일정한 수준으로 재생하고 농도를 지속적으로 유지한는데,

이러한 효소의 연속 재생은 효소가 소량으로도 우리몸에 매우 집중적으로 사용되는 것을 의미한다. 

인간의 몸이 효소 시스템안에서 자체 몸무게를 변경 또는 유지되는 것 또한 효소의 연속 재생의 원리이다.


효소는 신호 전달 및 세포 조절등 생명체 내부에서 다양한 기능한다.

(또한 감염 세포에 의해 바이러스에도 효소를 포함 할 수가 있다 : HIV, 인플루엔자 바이러스 등)


효소의 중요한 기능은 동물의 소화 시스템에 있다.

아밀라제 및 프로테아제 ( 당분 또는 단백질 )분해하여 창자 흡수 될 수 있다.


당 분자(가수분해 하여 소장 에서 흡수): 말 토스, 글루코스.

장내 미생물 분해: 셀룰로오스 분해 셀룰라제 효소.

당분의 대사: 당분 해 효소.


효소는 경로에서 발생하는 다음 단계를  결정한다.

효소가 없으면, 신진 대사나 세포의 요구에 부응 할만큼 빠르게 반능하지 못한다.


예를 들어, 포도당은 하나 이상의 탄소를 인산화하기 위해 직접 ATP와 반응 할 수있다.

효소가 없으면 이런 반응은 아주 천천히 미미하게 발생한다.


헥소 키나제를 첨가하는 경우에는, 이러한 느린 반응은 6의 인산화가 너무 급격히 즉, 발생하는 숯을 제외 일어날 계속 혼합물을 짧은 시간 후에 시험하는 경우, 글루코스 -6 - 포스페이트는 단지 중요한 제품이 발견된다.


네바다 대학의 실험에 의하면,

효소의 최적 흡수도는

온도 24 C

PH 6( 최 약산성)에서 가장 높은것으로 실험되었다.

Enzymes are large biological molecules responsible for the thousands of metabolic processes that sustain life.


They are highly selective catalysts, greatly accelerating both the rate and specificity of metabolic reactions, from the digestion of food to the synthesis of DNA. 


Most enzymes are proteins, although some catalytic RNA molecules have been identified. 


Enzymes may employ organic (e.g. biotin) and inorganic (e.g. magnesium ion) cofactors to assist in catalysis.

Almost all chemical reactions in a biological cell need enzymes in order to occur 

at rates sufficient for life. 

Most enzyme reaction rates are millions of times faster than those of comparable un-catalyzed reactions. 


As with all catalysts, enzymes are not consumed by the reactions they catalyze, nor do they alter the equilibrium of these reactions. 

However, enzymes do differ from most other catalysts in that they are highly specific for their substrates. 

Enzymes are known to catalyze about 4,000 bio-chemical reactions.


Enzyme activity can be affected by other molecules.

 Inhibitors are molecules that decrease enzyme activity;

activators are molecules that increase activity. 

Many drugs and poisons are enzyme inhibitors. 


Activity is also affected by temperature, pressure, chemical environment (e.g., pH), and the concentration of substrate. 

Some enzymes are used commercially, for example, in the synthesis of antibiotics.  

some household products use enzymes to speed up biochemical reactions (e.g., enzymes in biological washing powders break down protein or fat stains on clothes; 

enzymes in meat tenderizers break down proteins into smaller molecules, making the meat easier to chew).


 Coenzymes transport chemical groups from one enzyme to another.

Some of these chemicals such as riboflavin, thiamine and folic acid are vitamins (compounds that cannot be synthesized by the body and must be acquired from the diet). 


Enzymes serve a wide variety of functions inside living organisms. 

They are indispensable for signal transduction and cell regulation, often via kinases and phosphatases.


Viruses can also contain enzymes for infecting cells,

 such as the HIV integrase and reverse transcriptase, or for viral release from cells, like the influenza virus neuraminidase.


An important function of enzymes is in the digestive systems of animals. 

Enzymes such as amylases and proteases break down large molecules (starch or proteins, respectively) into smaller ones, so they can be absorbed by the intestines. 


Starch molecules, for example, are too large to be absorbed from the intestine, but enzymes hydrolyze the starch chains into smaller molecules such as maltose and eventually glucose, which can then be absorbed. 

Different enzymes digest different food substances. In ruminants, which have herbivorous diets, microorganisms in the gut produce another enzyme, cellulase, to break down the cellulose cell walls of plant fiber.


Enzymes determine what steps occur in these pathways. 

Without enzymes, metabolism would neither progress through the same steps nor be fast enough to serve the needs of the cell.


 Indeed, a metabolic pathway such as glycolysis could not exist independently of enzymes. 

Glucose, for example, can react directly with Adenosine Triphosphate Adenosine triphosphate (ATP)

 to become phosphorylated at one or more of its carbons.

 In the absence of enzymes, this occurs so slowly as to be insignificant.


 However, if hexokinase is added, these slow reactions continue to take place except that phosphorylation at carbon 6 occurs so rapidly that, if the mixture is tested a short time later, glucose-6-phosphate is found to be the only significant product.

Exercise A (enzymes)

Part 1 of Exercise A investigates the effect of varying temperature on enzyme activity. 

It's important to remember that the increased absorbance means greater amount of product and a higher reaction rate. Table 10A-1 is a summary of the investigation results:

The greatest activity vs time is displayed from the enzyme at 24 degrees Celsius. At 50 degrees the enzyme is most likely denatured and at 10 degrees it will work for a short time but at a much lower activity as compared to 24 degrees.


Part 2 further investigates the varying effects of external forces on enzyme activity. This time we examine pH. The results are listed below:

The results show that the best pH for our enzyme is somewhere around 6. At other concentrations of hydrogen ions it will not function properly.
